Title: BLADE Directed Evolution of SaCas9 gRNAs Improves Gene Editing Efficiency

Description:

CRISPR-Cas9 efficiency is often limited by misfolding between the sgRNA scaffold and spacer regions. Using the BLADE-SELEX directed evolution platform, we isolate hundreds of diverse gRNA scaffolds from an initial library of over 100 trillion variants. Scaffolds variants improve predicted folding, boost editing efficiency across all tested sites, and open new research avenues for CRISPR development and large-scale data generation.
