Modulation of the backbone structures of oligonucleotides has a profound impact on the pharmacokinetics and pharmacodynamics of RNA medicines. In the siRNA therapeutics field today, chemically stabilized synthetic RNA, while preserving essential interactions between siRNA molecules and RISC-associated proteins, is critical for ensuring sustained therapeutic effects. In this webinar, we will discuss how subtle structural modifications influence the oligonucleotide’s structural behavior and the biological properties. We will also highlight our recent advances in siRNA chemistry using extended nucleic acid (exNA), a platform featuring a single methylene insertion between the internucleotide phosphate and 5′-position of canonical nucleic acids.
