Title: Selection of DNA Aptamers to Inhibit TDP-43 Aggregation in ALS

Description:

Amyotrophic Lateral Sclerosis (ALS) is a fatal neurodegenerative disease characterized by the loss of motor neurons. With an unknown pathology in its sporadic form, current hypotheses suggest the cytoplasmic mislocalization and subsequent aggregation of TAR-DNA Binding Protein-43 (TDP-43) may play a significant role in the neuronal toxicity associated with ALS. Our work aims to develop new DNA aptamers capable of inhibiting TDP-43 aggregation, where the top candidates will be evaluated for their effects on cellular toxicity, in vitro aggregation, and cytoplasmic mislocalization. We are keen to understand if DNA aptamers are suitable aggregation inhibitors in vitro for ALS, and if this inhibition translates to a reduction in neuronal toxicity.
